Study of a method of B and F separation of hCG by radioreceptor assay

In the radioreceptor assay, one of the key steps is the separation of bound radio substance (B) and free radio substance (F) at the end of the reaction. We chose polyethylene glycol 6000 (PEG) and .gamma.-globulin protein for the separation of B and F. The concentration of PEG for optimal precipitation of receptor bound hCG was 7% (W/V) 0.42% of .gamma.-globulin protein got the good result of separation. Comparison with methods of millipore filter and high speed centrifuge revealed that the nonspecific binding was highest among the three methods when we used millipore filter. The results were influenced by .gamma.-globulin protein from different sources. The .gamma.-globulin protein from SIGMA was better than that prepared by our laboratory. Comparison between .gamma.-globulin protein and serum showed that with the former, the binding was 25.9%, and with the latter 12.6%. This is a good method for separation of B and F, because it is convenient and does not need specific equipment.
