Binding studies on the native and heat-denatured DNA with manganese (II) ions by the equilibrium dialysis (temp. 4.degree.C, pH .apprxeq. 6, equilibration time 70 h) in various ionic strengths (0.01-0.1) has been performed. The concave-downward Scatchard plots obtained at various ionic strengths, an insusceptibility of curvilinear isotherms against the graphical break-down and rectilinear run of isotherms for the heat-denatured DNA complexes with manganese (II) ions.sbd.ruled out an occurrence of intercalation of manganese (II) ions in the interstrand sites within DNA. From the Record et al. (14) plot and experimentally obtained binding constants, Kobsd, the stoichiometry binding of manganese (II) ions to phosphate residues of DNA (1:1) has been determined.
