Natural plant enzyme inhibitors: comparison of the action of tuber protease inhibitors on animal pancreatic systems

The inhibition of total proteolytic (caseinolytic), tryptic (benzoyl arginine p-nitroanilide hydrolysis) and chymotryptic (acetyltyrosine ethyl ester hydrolysis) activities in nine different pancreatic preparations by potato, arrow root and colocasia tuber inhibitors was compared. Potato inhibited the caseinolytic activities of rabbit and horse pancreatic systems maximally (32.6 units/mg protein) whereas it had the least effect on the human preparation (4.16). Its inhibition of the tryptic activity of rabbit system (142) and chymotryptic activities of equine (61.1) and human (54.2) pancreas was also maximum. Arrow root inhibitor was most effective (15.6) in blocking porcine tryptic action. Equine chymotrypsin was more susceptible to the three tuber inhibitors compared to the other animal chymotrypsins. Partial inhibition of cat tryptic activity by potato and arrow root inhibitors and of rat chymotryptic activity by potato suggest the presence of isoenzymes differing in susceptibility to inhibitor action.
