Acetaldehyde, the proximal metabolite of ethanol was administered into pregnant CF rats i.p. in single and triple doses (50, 75 and 100 mg/kg) on days 10, 11, and 12 of gestation. Fetuses which were collected on day 21 of gestation along with the placentae presented malformations akin to those of fetal alcohol syndrome. Histological sections of term placentae revealed thick white patches, thickened placental barrier, perivascular fibrosis and distorted labyrinthine architecture. Altered placental structure suggests the involvement of placenta in the pathogenesis of the acetaldehyde teratogenicity.
