Production of the leukocyte inhibitory factor (LIF) by peripheral blood lymphocytes in vitro and LIF-like plasma activity in paraproteinemic patients

Patients affected by multiple myeloma or Waldenstrom's disease have a complex series of immunological defects, however, the abnormalities of granulocytic function and the alterations of lymphokine synthesis in these patients are poorly understood. In 6 paraproteinemic patients the following were evaluated: the polymorphonuclear granulocyte (PMN) random migration in normal or paraproteinemic plasma; the synthesis of leukocyte inhibitory factor (LIF) in vitro by peripheral blood lymphocytes stimulated with concanavalin A and the PMN random migration in LIF-containing medium. PMN from paraproteinemic patients have a random migration pattern comparable to controls, in nomral or paraproteinemic plasma and in LIF-containing medium. Plasma from 2 IgA myeloma patients contained LIF-like factor(s) with strong migration inhibitory activity. Peripheral blood lymphocytes from paraproteinemic patients and from normal controls synthesize comparable amounts of LIF or LIF-like factors when stimulated in vitro by concanavalin A.
