Serum is placed in a well in agarose gel containing guinea pig complement and J-negative bovine erythrocytes coated with lipopolysaccharide of Brucella abortus biotype 1. After incubation, a positive serum produces zones of haemolysis. This test was compared with established serological tests in serum samples from 1041 brucella-free cattle, 51 infected with B. abortus biotype 1 or 4, and from 6 heifers vaccinated with strain 19. It was more sensitive than the other tests for detecting biotype 1 infection, but less sensitive in biotype 4 infection. It became positive later in vaccinated heifers, and a positive response persisted for nearly 2 years.
