Density gradient fractionation of antibody forming cells

Isolation of enriched populations of antibody forming cells (AFC) can facilitate their comprehensive surface characterization and may also help in the generation of immunosuppressive-idiotypic sera. Such an attempt was made by centrifuging immune lymphocytes on a discontinous density gradient of Percoll. IgM and IgG antibody producing cells were assayed by plaque formation. On centrifugation at 400 g for 20 min, it was observed that the IgM-AFC were concentrated in fractions of buoyant density 1.065 to 1.071 g cm-3. IgG-AFC were uniformly distributed. Percoll density gradient may be useful in obtaining relatively enriched populations of IgM antibody secreting cells.
