The compound structure of the R allele in maize

The stippled-aleurone allele (Rst) mutates to self-coloured (Rsc) and to near-colourless r (nc) aleurone. Heterozygous combinations involving several r (nc) mutants and Rsc alleles were tested for resynthesis of the Rst allele. Resynthesis was successful in heterozygous combinations of Rsc alleles, either paramutagenic or nonparamuta-genic, with a near-colourless aleurone mutant rs (nc) 1-3. Heterozygous combinations of Rsc with rs (nc) 1-2 and with rr (nc) mutants did not yield resynthesized Rst alleles. Results indicated that the stippled phenotype is dependent on at least two R locus components separable by crossing over. One component is the structural gene for self-coloured aleurone Sc and the second is an inhibitor IT of Sc. The stippled phenotype is produced when Is and Sc are brought together in the cis configuration by a cross over.
