Effect of oxotremorine on isolated guinea pig vas deferens

Oxotremorine, the tremorogenic agent, produced a concentration dependent potentiation of responses of the isolated guinea pig vas deferens to hypogastric nerve stimulation and transmural stimulation. When the facilitatory effect of oxotremorine was at a maximum, the responses to exogenous epinephrine were also augmented. Partial tachyphylaxis developed to the potentiating effect of oxotremorine. The effect of oxotremorine was reduced in reserpinized preparations and in presence of hemicholinium 3. The effect of oxotremorine on evoked responses of guinea pig vas deferens is mediated through an indirect mechanism of release of norepinephrine from the neural tissue; this release is cholinergically mediated.
