Chlorambucil-induced changes in chromatin methylation and their modulation by sodium butyrate

Chlorambucil treatment for 2 h increased [rat Walker 256 tumor cell] chromatin methylation in sensitive cells but not in alkylating agent-resistant cells. Longer exposure to chlorambucil resulted in the inhibition of methylation in both cell lines. Pretreatment of cells with the transcriptional inducer sodium butyrate abolished the initial stimulation and potentiated the inhibition of methylation. These changes in DNA and nuclear protein methylation correlated directly with the loss of condensed chromatin previously observed following alkylating agent treatment.
