Biochemical studies of erythrocytes isolated from patients with myoglobinuria

A close correlation of pathogenesis between malignant hyperthermia and myoglobinuria, which developed during operation under general anesthesia was previously reported. Myoglobinuria might be a model to study the cause of malignant hyperthermia. In addition many researchers have paid attention to the usefulness of erythrocytes to study the pathogenesis of muscle disease, because they consider some muscle diseases as a systemic membrane disease. A comparison of the erythrocyte osmotic fragility between normal adult and myoglobinuria patients was made by the coil planet centrifuge system (CPC). By comparison with some conventional methods of routine fragility test, CPC was found to be a precise and highly reproducible method for analyzing the osmotic fragility of erythrocytes with a small amount of blood. There was apparently an increase of erythrocyte osmotic fragility in myoglobinuria patients by aging at 37.degree. C, while no differences were found between 2 groups incubated at 0.degree. C, on the other hand. Na-K ATPase activity of erythrocyte ghosts of the patients showed a fewfold elevation, and chemical analysis of phospholipid and protein components extracted from erythrocyte ghosts showed no significant difference. The elevation of Na-K ATPase activity of erythrocyte membrane of myoglobinuria would suggest the abnormality of erythrocyte membrane, and the same abnormality would also be suspected in the muscle cell membrane.
