Effect of hydroxyurea on UV-induced mitotic crossingover & gene conversion in Saccharomyces cerevisiae

A study on the mechanism of UV-induced mitotic crossing over and gene conversion was made with stationary phase cells of strain D7 of S. cerevisiae. Incubation of cells in liquid synthetic complete medium containing 40 mM of hydroxyurea (HU) for > 8 h increased the frequency of convertants whereas survival and mitotic crossing over were not affected. Incubation of UV-irradiated cells with HU also increased the frequency of gene convertants with no measurable effects on survival and mitotic crossing over. The frequency of UV-induced gene convertants increased if HU was added within 4 h after UV-irradiation. UV light induces mitotic crossing over and gene conversion by bringing about pairing of homologous chromosomes and the mechanisms are similar for both the events.
