Direct demonstration of the virus in enzootic bovine leukemia using cell cultures

Bovine oncovirus was demonstrated in bovine peripheral blood lymphocytes with the syncytial infectivity assay (SIA) induced in a permanent cell line CC81 (cat cells transformed by murine sarcoma virus). The technique included co-cultivation on a cover slip of blood lymphocytes maintained in Eagle's medium together with CC81 grown in minimal essential medium supplemented with 10% serum free of antibodies. After 5, 7 and 9 days haematoxylin-eosin stained cultures were examined for syncytial growth. In positive cases syncytial formation occupied 5-100% of the monolayer. The syncytial phenomenon was suppressed or abolished when antiserum to bovine oncovirus was added. Of 12 cattle positive to the immunodiffusion test 10 reacted to SIA, as did 2 out of 38 AGID-negative cattle.
