Methods for isolation and determination of medium-chain fatty acyl thioesterase (thioesterase II) from lactating rat mammary gland are described. Thioesterase II is purified from mammary cytosol using DEAE-cellulose and Ultrogel AcA54 chromatography. Thioesterase II was not recognised by antibodies to fatty acid synthase or thioesterase I, but thioesterase II antibodies inhibited its activity. Other properties of thioesterase II which are discussed are physicochemical properties, amino acid composition, substrate specificity, kinetic properties and inhibition.
