Clinical pharmacokinetics of a microcrystalline theophylline

Eighteen non-smoking asthmatic patients received a dose of microcrystalling theophylline . The maximal serum concentration is reached in a short time, independant of the dose, 81 min with a dose < 5 mg/kg (10 patients), 78 min with a dose > 5 mg/kg (8 patients). The micronization enhances theophylline absorption, reducing gastrointestinal irritation and allowing faster efficient serum levels. The volume of distribution, half-life and clearance values obtained allow an individual dosage regimen to maintain a 10 mg/l serum level. The validity of this regimen, studied in 13 patients, was satisfactory.
