An experimental animal model for in vitro evaluation of sensitization was developed by determining the specific and comparatively nonspecific histamine secretion by animal cells. Sections of the lung tissue from sensitized or nonsensitized animals were acted upon with different factors, and histamine was determined by fluorimetry. A state of sensitization to horse serum and aggregated human Ig was elicited in guinea pigs, histamine secretion in vitro being detected. Various factors were tested for their effect on the process. Experiments were extended to a model of anaphylactic shock, which may occur upon 1st contact of the animal with antigen. Histamine secretion by lung sections of nonsensitized guinea pigs in the presence of aggregated human Ig could not be found if own animal serum was the source of complement. Histamine secretion was detected when rat serum was applied. The results obtained extend possibilities of safety testing of human Ig preparations.
