Evaluation of the metabolic action of glucagon in subjects undergoing anti-insulin therapy

Eight obese male subjects were studied to see whether the lipolytic action of glucagon combined with the contemporary block of insulin release is effective. Glucagon (1 mg/kg of body wt per min) was injected and blood glucose, total lipids, triglycerides, cholesterol, free fatty acids, glycerol, immunoreactive insulin, growth hormone, gastrin and prolactin were measured. The same test was repeated after pretreatment with diphenylhydantoin (4 subjects) or diazoxide (4 subjects). There was an increase of blood glucose and immunoreactive insulin after glucagon administration. Diazoxide and diphenylhydantoin produced an augmented blood glucose increase and lower levels of insulin both in basal conditions and in response to the stimulus. The therapeutic use of these drugs is suggested to reduce hyperinsulinemia in obese subjects.
