The bromelain potencies of commercial bromelain tablets (preparation 1, 2 and 3) were measured. The bromelain potencies of preparation 1, 2 and 3 which were extracted with aqueous solution containing 0.03 M cysteine and 6 mM EDTA (pH 4.5) (solution A) were 20.8, 21.5 and 94.9% of labeled potency, respectively; the potencies with the same aqueous solution containing 0.4 M NaCl (solution B) were 80.1, 74.6 and 98.1%, respectively. Polyacrylamide gel electrophoresis at pH 4 showed that more amounts of basic protein(s) were extracted from preparation 1 with solution B than with solution A. In the preparations which have shown lower potency as extracted with solution A, bromelain may be adsorbed by the insoluble excipient or may be decreased in solubility due to the presence of excipient and other additives.
