Asynchrony in proliferation of sister nuclei in colcemid-induced polykaryons of muntjac lymphocytes

The effects of prolonged exposure to colcemid on the lymphocytes of barking deer M. muntjak (2n = 6.female. .+-./7.male.) were examined. The treatment cause formation of hyperdiploid cells having 6-7 micronuclei. Certain polykaryons exhibited asynchrony in their proliferation kinetics leading to premature condensation of the chromatin of the lagging nucleus. The late-S prematurely condensed-chromosomes displayed a banded appearance corresponding to their replicating sites. Among binucleate interphases, nuclei in different phases of cell cycle were observed. The asynchrony of proliferation was explained on the basis of hindrance in cellular transport of inducer metabolites due to annihilation of microtubular assembly by colcemid.
