Concentration of mast cells was estimated, and their distribution noted, in different organs of rats rendered hyperglycemic with alloxan. Increased proliferation of mast cells was noted in pancreas, thyroid, skin and mesentery after induction of diabetes. Pretreatment with insulin designed to improve the diabetic state by lowering hyperglycemia reduced the diffusely distributed mast cells in pancreatic islets, skin and mesentery. With few exceptions, slight increases in the concentration of mast cells were also noted in tongue, stomach and skeletal muscle in the pronounced hyperglycemic state. The increase in mast cells in the islets of Langerhans, skin and other tissues apparently was not due to the alloxan toxicity only, but was brought about as a consequence of diabetic hyperglycemia.
