An optimized lead capture electronhistochemical technique for the demonstration of lysosomal and non lysosomal acid phosphatase activity in nervous tissue

The problems inherent in electron histochemistry are outlined with particular reference to acid phosphatase. An ultrahistochemical technique was optimized for demonstration of this enzyme in nervous tissue with regard to fixation, tissue preparation and incubation medium components. Glutaraldehyde fixation (2 h), 50 .mu.m sections from a modified vibratome and high levels of substrate and Pb ions give best results. Using the optimized technique an unusual .beta.-glycerophosphate-hydrolyzing non-lysosomal acid phosphatase is described associated with spinal cord nerve terminals and the Golgi of their cell bodies in dorsal root ganglia.
