Hydrolysis of procaine in liposomal suspension

Based on the partition model, the base-catalyzed hydrolysis of procaine was examined in liposomal suspension prepared from phosphatidylcholine and compared to its spontaneous hydrolysis in aqueous solution. Rate retardation of the hydrolysis was seen for the free base, but no interaction was found between the protonated form and liposomes. The hydrolysis rate of the free base was reduced by 65-85% in the lipid phase with the partition coefficient .apprx. 190. When cholesterol was incorporated in the liposome, a slight rate enhancement was seen compared with the cholesterol free case.
