Isolation and functional maintenance in culture of syncytiotrophoblasts from human placenta

A method is described for obtaining pure preparations of syncytiotrophoblasts from human term and subtern placentae. Cells prepared by this method give immunofluorescent reactions for trophoblast specific proteins-hCGP1. Isolated syncytiotrophoblasts were maintained in culture in morphologically healthy state for several months. Subterm and term syncytiotrophoblasts remain metabolically functional in vitro and secrete hCG at rates proportional to their synthetic performance in vivo. Replication of the cells during the period of culture is undetectable by 2 criteria: measurement of incorporation of radioactive thymidine and analysis of the DNA content of the cultured cells.
