Cytogenetic studies in leukemias--blastic crisis of chronic myeloid leukemia

Cytogenetic studies were carried out during the blastic crisis of chronic myeloid leukemia patients. In addition to metaphases with Philadelphia (Ph1) chromosomes, other abnormalities were observed. These included monosomy of E, trisomy of G and trisomy of F group chromosomes. In 50% of the patients, 2 Ph1 chromosomes were observed. In 1 patient with extramedullary blast crisis, a separate cell line was found at the extramedullary site while in 2 other patients, identical cell lines were found in the bone marrow and extramedullary sites. A non-random evolution of karyotypes during progression of the disease was shown. The acquisition of a 2nd Ph1 chromosome seems to occur in many patients later during the progression before blastic transformation.
