Effect of storage, different media constituents and dilution of axenic medium on the growth of axenic culture of Entamoeba histolytica

Modified TP-S-1 axenic medium (prepared without ascorbic acid but containing 0.2 or 0.3% L-cysteine) supports the growth of axenic E. histolytic (strain STA-CDRI) even after storage for 3 wk at 4.degree. C. When cysteine solution stored for 2 and 3 wk was used to prepare the axenic medium, 0.3% L-cysteine concentration supported growth, whereas it was poor with 0.1%. Removal of trypticase or vitamin mixture from axenic medium prepared with 0.2% L-cysteine reduced the growth of amoebae by 50%. Medium prepared without serum or panmede failed completely to support the growth of amoebae. Dilution of axenic medium in ratio of 1:2 and 1:3 with phosphate buffer saline (containing 0.2% L-cysteine, pH 7.0), resulted in complete lysis of amoebae in 6-9 days.
