Cultivation of axenic Entamoeba histolytica from small inocula

Under suitable conditions of stable negative O-R [oxidation-reduction] potential produced by 0.05, 0.1, 0.2 and 0.3% L-cysteine in modified TP-S-1 Diamond's medium, axenically grown E. histolytica (strain NIH-200) could be maintained in subculture starting from 15-250 trophozoites/ml. of the medium. Maximum growth of amoebae was reached earlier when the medium was replaced after 6-8 days. A standard inoculum of 250 amoebae/ml. showed 800-1200 times growth in 2 wk. By reducing inoculum of amoebae to 250/ml. the subcultures of axenic amoebae can be made after 2 wk which was not possible before.
