Induction of dominant lethal mutations in mice with chlordiazepoxide hydrochloride

The genetic damage induced by the tranquilizer, chlordiazepoxide hydrochloride was studied in male germ line of CBA strain in mice. Two concentrations of the drug (0.8% and 1.6%) were dissolved in distilled water and given as drinking water for 4 wk to adult male mice which were then continuously mated with a series of virgin females of the same strain. The incidence of intrauterine deaths was taken as the criterion for assessing genetic damage. The stages corresponding to secondary spermatocytes and spermatogonia were found to be sensitive for both doses. The combined incidence of intrauterine deaths for the 4 wk was 8.93% and 11.4% respectively, for the 2 concentrations which recorded a significant increase over the control values.
