Method of regressive staining of semi-thin sections

The method presented permits well-stained semi-thin sections of material embedded in epoxy resins (Epon 812, Araldite, Westopal) to be obtained quickly. These sections serve for the preliminary, light microscopic assessment of material intended for further processing and analysis in the transmission electron microscope. Staining with this method should proceed in a dye or a mixture of assorted dyes on a watch-glass at room temperature or 40.degree. C. When the sections are rinsed with distilled water and adhere to the slide they should be subjected to the process of differentiation at a higher temperature (40-60.degree. C). The cedar oil medium provided by dropping the oil on the sample markedly induces the differentiation time.
