Effect of the analogue of cinnarizine AS2 on the cardio-vascular system

The effect of the analogue of cinnarizine, AS2 (N'-benzhydril, N"-allyl-piperazine dihydrochloride), was examined on the cardiovascular system in cats. Its effect was studied on arterial pressure, minute and stroke volume, general peripheral resistance and the function of the left ventricle. A 2nd series of experiments were carried out to study the effect of AS2 on coronary blood flow and the O2 metabolic processes in the myocardium. Cinnarizine is tested as control. In the doses applied AS2 demonstrated a very pronounced coronarodilatating activity, its effect being twice stronger compared with that of cinnarizine. Both drugs lead to decrease in the general peripheral resistance and the work of the right ventricle. The established coronarodilatating activity of the compound is probably due both to a direct effect on the smooth-muscle elements of the vascular wall and to a decrease in the O2 uptake by the myocardium.
