In vitro stimulation of human breast tissue by human prolactin

Normal mammary tissue from 30 women was cultured for 4 days in Waymouth's MB 752/1 medium with no hormones (control), insulin (I), I + ovine prolactin (OP), I + human prolactin (HP) or I + human placental lactogen; all hormones were used at 5 mu g/ml medium. 8 h before the end of culture, 0.1 mu g colchicine/ml or 1 mu Ci [3H]thymidine/ml was added to each culture. There was little mitosis or DNA synthesis in the absence of hormones in vitro although the tissue appeared healthy. Addition of I caused an increase in mitotic activity of 390% above the control group. I + OP produced only a minimal increase above that obtained with I alone. However, I + HP or I + placental lactogen resulted in a 36% increase in mitotic activity above that of the group with only I.
