Orally given kinases were found to induce fibrinolytic and clinical effects similar to those following parenterally administered enzymes.' The absence of side effects (pyrogenicity, chills, hypotension, hemorrhage) characterizing parenteral routes of enzyme administration denoted little, if any, systemic absorption.2 The possibility was considered, therefore, that orally given kinases might catalyze the formation of plasminogen activator from a zymogen present in gastric juice. This would imply that the administered kinase is not absorbed per se, but participates in the endogenous formation and absorption of plasminogen activator. Experiments described herein suggest the feasibility of employing streptokinase orally as the pharmacologic means for releasing endogenous plasminogen activator into the circulation. Indeed, animal and clinical studies now in progress document fibrinolytic and anti-inflammatory effects following orally given streptokinase (Varidase).
