The separation of crude polysaccharides from the culture filtrates of H. [Phialophora] pedrosoi, H. compactum [P. compacta] and H. [P.] dermatitidis was effected by ethanol precipitation after dialysis and concentration. Fractionation by gel filtration on Sephadex G-75 column gave in each case galactomannan and glucan, the former being further fractionated into 1 major and 2 minor subfractions by DEAE Sephadex chromatography. All the major fractions were homogeneous and had strong precipitating activities against homologous anti-whole rabbit sera. In agar gel double diffusion, 6 galactomannans isolated from each str. gave a single precipitin arc which fused completely with adjacent ones. In the precipitin reaction these polysaccharides were also highly cross reactive to heterologous anti-P. sera. However, no serological activity was observed on any of the glucan fractions purified by a method similar to that used for the galactomannans.
