Intensity of lipid biosynthesis in experimental ischemia

Ischaemia of the myocardium was induced by ligating the posterior branch of the left coronary artery of 9 rabbits. After 5 days some of the rabbits were given by vein sodium acetate-2-14C, 60 mu Ci/kg bodyweight, and blood was taken after 60 min. When the rabbits were killed, lipids in serum, heart and liver were extracted. In another experiment tissue slices from 5 normal and 4 ischaemic rabbits were incubated for 60 min in a medium with sodium acetate-2-14C abd glucose-1,6-14C after which lipids were extracted.In the normal rabbits there were 4 forms of lipoprotein, alpha - and beta -lipoproteins, subchylomicrons and chylomicrons; these forms were preserved during myocardial ischaemia. Radioactivity of serum proteins during infarction of the myocardium fell to half of that of the normal rabbits but that of lipoproteins rose. In the test rabbits incorporation of 14C into cholesterol, triglycerides and fatty acids rose while that into lecithin, phosphatidyl serine and sphingosine phosphatides of liver fell. Changes in lipid metabolism in vivo were similar to those in vitro. The relation between lipid changes in acute myocardial infarction and other accompanying disorders of myocardial functions are discussed.
