The effect of synthetic estrogens (mestranol, estrone cyanate and quinestrol) on carbohydrate and lipid metabolism during menopause

The effects of mestranol, oestrone cyanate and quinoestrol on glucose and free fatty acids in blood of 65 healthy postmenopausal women were studied in insulin tolerance tests. After 80 mu g mestranol daily for 10 days fasting values rose but had returned to initial values after 6 such periods of treatment. Insulin resistance due to mestranol was more distinct with 0.025 IU insulin/kg bodyweight than with 0.05 IU. The metabolic effects of mestranol were fully reversible. Only after the first period of treatment, 0.2 mg daily for 10 days, were fasting glucose and free fatty acid values increased by oestrone cyanate. Quinoestrol, 1 mg every 3 weeks, increased fasting glucose values and after 6 doses entry of glucose into the blood was considerably retarded in the insulin tolerance test.
