Sixty-three male Wistar rats weighing 150 g were fed for 3 weeks on laboratory chow without or with 2% oxonic acid (a potent inhibitor of hepatic uricase) alone or with 3% uric acid. Rats given oxonic acid developed hyperuricaemia and uricosuria; their weight gain was reduced, but significant weight loss or anorexia did not occur. All hyperuricaemic rats developed nephropathy; none developed arthritis. The rats are considered to be a suitable model for the study of hyperuricaemia and nephropathy, comparable to human disease in which excess uric acid is produced.
