The influences produced by treatment with SO2-water, running water, acidified alcohol and a graded series of ethanol at different temperatures on the fluorescence intensity of the nuclei stained by auramine-SO2 were studied. The treatment with SO2-water at room temperature results in the extraction of a specifically bound dye, therefore the treatment with SO2-water should be performed in refrigerator at 2-5.degree. C. The differentiation of stained preparations should not be made using acidified alcohol. The treatment of stained preparations by alcohol-acid solution presumably reveal some fraction of aldehyde groups of hydrolyzed DNA. The next conditions are recommended to carry out at differentiation of the Feulgen reaction with auramine-SO2: the treatment with SO2-water at 2-5.degree. C in 3 changes/3-5 min in each; washing in running water for 15-20 min, the whole treatment in a graded series of ethanol for 30-60 min, followed by the air drying of preparations in the dark; mounting in acryl or nonfluorescence vaseline oil immediately before measuring.
