Finding optimal conditions for sample preparation for the determination of NH3 in the rumen juice of cattle

Rumen fluid was sampled by naso-oesophageal tube from 10 cattle and ammonia was estimated photometrically with dipotassium tetraiodomercurate as reagent after storage for 0 to 30 h at 0 degrees , 20 degrees , 30 degrees or 37 degrees C. Values increased with increasing storage time and temperature. For true NH3 content immediate estimation, avoiding a protracted distillation or microdiffusion method, was recommended or, failing that, storage at under 0 degrees and direct estimation immediately after thawing.
