Total adenosine triphosphatase (ATPase) activity of erythrocyte membranes of young calves increased during anaplasmosis, due primarily to tipling of Mg-dependent ATPase (oubain-insensitive) activity at the initial detection of Anaplasma in erythrocytes and at the peak of parasitaemia. The Na, K-dependent ATPase (ouabain-sensitive) activity did not change, neither did the pH for optimal activity of total ATPase. However, the Michaelis-Menten constant of ATP for the enzyme decreased. Substrate specificity of both components of total ATPase activity associated with erythrocyte membranes of diseased calves varied from that of erythrocyte membranes of normal calves. ATPase activity was not altered by experimentally induced anaemia comparable in severity to anaplasmosis. It was concluded that part of the increase in ATPase activity during anaplasmosis was contributed by the parasite.
