Evaluation of a modified Wickerham medium for identifying medically important yeasts

A procedure employing a modified Wickerham medium containing a pH indicator and solidified in individual agar slants was compared with the standard auxanographic technique and the oxidation-fermentation (O-F) method. Of 120 isolates 96% gave the expected assimilation patterns within 14 days using the modified Wickerham medium, and 91% agreement with expected results was obtained using the auxanographic technique. Only 70% of strs. tested yielded the expected pattern when the O-F medium was used. Further tests have shown the modified medium to be reliable, sensitive and easier to interpret than the standard Wickerham broth and auxanographic media. When conditions are carefully controlled, the medium can be sterilized by autoclaving, thus simplifying preparation.
