Activation of nisin production by UV-irradiation in a nisin-producing strain of Streptococcus lactis

A survival curve was prepared of 6-h broth cultures of Str. lactis-6, washed and suspended in phosphate buffer (0.02M, pH 7), and exposed to UV irradiation of 0-10 800 ergs/mm2. A dose of 9000 ergs/mm2 gave <1% survival when subsequently plated on yeast/dextrose agar and high nisin-producing mutants were isolated. For determination of nisin production, isolates were grown in yeast dextrose broth for 16 h at 30 deg C and nisin was determined by the method of Tramer & Fowler [DSA 27, 1212]. An increase in nisin production of 50 and 100% was evident after the 1st and 2nd UV irradiation treatments, after the 3rd treatment a downward trend was found, although the range in nisin production was greater than in the non-irradiated parent culture. The 1st and 2nd exposures to UV irradiation had no effect on acid production of the cultures, but the 3rd reduced it. Survival of Str. lactis was greater after the 3rd than after the 1st 2 exposures (5.2 vs. 0.17 and 0.26%).
