Octanoate metabolism in the isolated perfused rat liver. I. Methodology and preliminary results

1. Male Wistar rats weighing 180 to 240 g were kept on a laboratory diet and were deprived of food for 15 h before removal of the liver. The livers were perfused with 70 ml recycled perfusion liquid and 100 mu mol caprylate, without or with 10 mmol lactate/litre medium, was added. In the control studies there was an increase in total ketone bodies, mainly acetoacetate; lactate decreased but glucose and pyruvate did not change significantly. The addition of caprylate increased acetoacetate and beta -hydroxybutyrate; lactate, pyruvate and glucose did not change significantly from control values. The addition of caprylate and lactate increased ketone bodies slightly and lactate decreased and glucose increased faster. When caprylate and lactate were added to perfusion fluid of livers from fed rats the acetoacetate value was lower than in any other group; lactate fell and glucose rose compared with control values but not as much as in the starved group given caprylate and lactate.
