Gas-solid adsorption chromatographic determination of short-chain fatty acids in rumen fluid

A 2 m X 2 mm stainless steel column packed with uncoated Chromosorb 101 (60/80 mesh) is conditioned for 2 to 4 days at 225 deg . After sterile filtration 9 ml of rumen fluid is added to 1 ml internal standard solution, 6.5 g isocaproic acid/litre 3M H3PO4. Chromatography of volatile fatty acids (VFA) is carried out at 190 deg with nitrogen gas flow 24 cc/min and using a flame ionisation detector. Good separation of peaks is achieved and isocaproic acid is eluted after 6 min. Results are given of VFA analyses on rumen fluid of groups of heifers taken 6 h after feeding on oats, beet pulp and molasses, grass pellets and hay.
