Evaluation of thick smear, Knott and membrane filtration methods for demonstrating microfilariae in blood

An evaluation was made on the detection of Wuchereria bancrofti microfilariae in a 20 mm3 thick blood smear, one ml aliquots of venous blood examined by the Knott technique and one ml of venous blood examined by membrane filtration. 105 people were examined - 12 known Wuchereria bancrofti mirofilaria carriers, 73 symptomatic patients and 21 asymptomatic individuals who lived in the same households as patients in the other 2 groups. Microfilariae were detected in 20% of the individuals by thick smear and 27% by both Knott and membrane filtration methods. Only 16% of those with clinial filariasis showed microfilaraemia and in all cases greater numbers of mirofilariae were detected by membrane filtration than by the other two methods.
