Tissue distribution of chloroquine and hydroxychloroquine in rats

Chloroquine diphosphate and hydroxychloroquine sulfate were given orally to Wistar rats of both sexes with a stomach gauge at a dose of 40 mg/kg daily, for 2 wk. The distribution curves of the 2 substances were analogous. The highest distribution curves of the 2 substances were analogous. The highest concentrations were found in the suprarenal gland; lower concentrations were found in the spleen, liver, kidney, lung, soleus muscle; even lower concentrations were found in the heart, skin, gastrocnemius muscle and plasma. The tissue chloroquine and hydroxychloroquine concentrations were generally higher in the female rats. The hydroxychloroquine concentration was 2-5 times lower than that of chloroquine.
