Growth and cytogenetic properties of a human endometrial adenocarcinoma which was cultured in vitro were studied chronologically, and the development into an established cell line was carefully observed. The explant material, obtained from a 71-year-old female, was placed into culture on May 16, 1968. A majority of the cultured cells (HEC-1-A) have grown well without interruption. A group of cells (HEC-1-B), however, sustained a short stationary period between 135 and 190 culture days. A jig-saw puzzle-like cellular arrangement and piling-up tendency in the growing process were the typical type of cells. The cytology revealed anaplastic and pleomorphic features. Sub-line (HEC-1-B) disclosed some morphological alteration. The chromosomal distribution has been stable at the diploid range. However, the number of stem cells changed from 47 to 46 (HEC-1-A). A marker, which is the largest submetacentric chromosome, has been present since the beginning of culture. The stem cell of HEC-1-B was found to have 94 chromosomes and to be a duplicant of the original HEC-1 cell. HEC-1-A has constantly been transplantable to a hamster cheek pouch and reproducible to make the original adenocarcinoma. No differences were observed in the histology of the tumor of HEC-1-B.
