Staining mechanism of thioflavin T with special reference to the localization of amyloid

The analysis of the alkaline fluorochrome thioflavine T yielded 1 yellowish green, 3 blue, 1 bluish green and 1 red fluorescing component. The yellowish green and blue fractions are different in their polymerization degree. They may change into one another, whereby in vitro only a change from yellowish green into blue takes place while depending on the charge conditions a formation of the yellowish green component from the blue fraction is also possible in the tissue section. Thioflavine T strongly deposits on acid tissue structures, apart from secondary amyloid an assortment of representative comparable structures (mast cells, cartilage etc.) was studied. Moreover histochemical reactions for characterizing the corresponding radicals in the tissue were applied. The dye adsorption is mainly an electrostatical one. Fluorescence colour depends on the density of the dye molecules on the substratum. As to the amyloid diagnostics reference must be made to the unspecifity of the thioflavine T fluorochromizing. The alternately composed interfibrillar amyloid substance must be considered here as staining substratum.
