Phase Ii study of emetine (NSC-33669) in the treatment of solid tumors

Fifty-one patients with various advanced solid tumors were treated with subcutaneous injections of emetine at a dose level of either 1 mg/kg of body weight daily for 10 days or 1.5 mg/kg of body weight twice weekly for 5 weeks repeated at intervals of 3 weeks. The toxicity was moderate and reversible, consisting mainly of muscular weakness, local pain, and transient cardiac arrhythmias. No complete or partial regression of tumor was observed. Emetine at the dose level and schedule of administration used in this study appeared to be inactive in bronchogenic carcinoma and demon strated no response in eight patients with gastrointestinal carcinoma, six with breast carcinoma, nine with genitourinary carcinoma, six with gynecologic carcinoma, and six with miscellaneous malignancies.
