On the carcinogenic action of methyl- and ethyl-nitrosourea in rats after transplacental and postnatal application

The carcinogenic action of methyl- and ethyl-nitrosourea was compared after transplacental (group I), neonatal (group II), and postnatal (group III) application in hooded rats. Altogether 2938 tumors were induced in 1540 animals. The types of tumors of the nervous system differ significantly in the three groups. Polymorphous gliomas (astrocytomas grade II, oligodendrogliomas grade II, and glioblastomas) as well as sarcomas occurred in adult rats more often than after transplacental or neonatal induction of tumors. After application of MNU or ENU, respectively, in adult rats higher differentiated malignant neurinomas prevailed whereas in the other two groups undifferentiated tumors dominated. The fetal brain was more sensitive in relation to the carcinogenic action than that of adult animals. On the other hand extraneural tumors were more frequent in group III than in the two other ones. Possible relationships between the stage of organogenesis and carcinogenic action as well as the age specific tumor types of the nervous system in man and their consequences in preventive oncology were discussed.
