Role of EPHX genotype in the associations of smoking and diet with colorectal adenomas

385 patients with colorectal adenomas and 396 polyp-free controls were recruited between 1997 and 2000 among individuals attending 8 hospitals in the Netherlands. Blood samples were drawn from the participants and EPHX polymorphisms in exon 3 and 4 were determined. The distributions of EPHX exon 3 and 4 polymorphisms did not differ between cases and controls. Smoking increased the risk of developing colorectal adenomas. For exon 3, this risk was confined to those with the fast (YY, TH) genotype. There was no difference in risk between carriers of fast or slow variants of exon 4. Intake of fat and oil was also positively associated with adenomas.
