Ibuprofen pharmacokinetics in patients with liver cirrhosis

In 18 patients with liver cirrhosis the kinetics of Ibuprofen was studied after administration of 200 mg of the drug orally. It was found that the biological half time of the drug was in patients with liver cirrhosis 1.85±0.77 (x±SD) on the average, while in the group of the healthy controls it was 1.45±0.32 hour (p<0.05). The area under the curve of concentrations was in the group of liver cirrhosis significantly greater (107.90±74.50 μg/cm3 × h), than in the group of controls (71.39±19.65 μg/cm3 × h); (p<0.05). Thus the biological availability of Ibuprofen was in the patients 52% higher, on the average. The results show that in states of liver failure Ibuprofen biotransformation is impaired, and lower than usually doses should be given in such cases.
